The thing to know about the Ai Yori Aoshi anime compared to the manga is that the manga is actually accompanied by a pretty serious story. The anime, particularly the second season, likes to trim away all the serious moments in order to just be another cheerful harem affair.
The first season of the anime, while it did change some things, skip chapters, and generally condenses the story a bit, actually was a decent adaptation of the manga. If you were to start reading at Chapter 54, you would be able to continue on just fine. The same can not be said for Season 2, which abridges and changes events so much that they become extremely different from the manga.
So long as you pick up the manga after Season 1, you don’t need to read the Ai Yori Aoshi manga from the beginning, though you would have a more complete experience if you did.
